Abstract

Introduction: Despite extensive investigations, the main mechanisms underlying the development of neuropathic pain are still not fully understood. On the other hand, due to existence of the several mechanisms in this case there is no effective treatment for chronic neuropathic pain. So intensive research is being done for finding new, efficient analgesic drugs. The aim of this study was to evaluate the effect of Umbelliprenin on behavioral responses of and the expression of proinflammatory factor expression in the chronic constriction injury of the sciatic nerve (CCI) model of neuropathy in male rats. Method & Materials: 56 male Wistar rats (230±30) were used in this experimental study. Animals were divided into 7 groups (n=8); control, sham, CCI and 3 Umbelliprenin groups with doses of 25, 50 and 100 microgram/rat. CCI model of neuropathic pain was used to induce chronic neuropathic pain. Umbelliprenin was injected from day before to 2 days after the surgery (intrathecally). Mechanical and thermal allodynia and thermal hyperalgesia were evaluated by von-Frey, acetone drop and hot-plate tests respectively, on day before the surgery and post-operative days (POD) 2, 4, 7, 10 and 14. Lumbar enlargement of spinal cord were collected for protein content and gene expression analysis. Results: The 6 (time) * 7 (treatment) mixed model ANOVA with Bonferroni indicated that allodynia increased in CCI group from POD 4 and hyperalgesia from POD 2 significantly in compared to the control group (P<0.05). Furthermore, Umbelliprenin 50 (P<0.01) and 100 (P<0.001) μg/rat significantly attenuated the induction of allodynia and hyperalgesia in compare to CCI group, and this effect was found to be significant from POD 7-14. One-way ANOVA indicated that IT injection of Umbelliprenin was able to reduce amount of TNFα protein content and IL-1β gene expression. Discussion: The results show that pre-emptive administration of Umbelliprenin before injury can prevent or delay the onset of neuropathic pain symptoms. Further more, it can reduce cytokine expression and protein content in lumbar spine of CCI rats. Our view is that these results could be useful in finding new drugs to treat or alleviate the symptoms of neuropathic pain.
