Abstract

Introduction: Neuropathic pain is a chronic pain caused by the defect or damage to the central nervous system or peripheral nervous system. Since chronic pains, including neuropathic pain, have an important effect on the quality of life of patients, it is considered necessary to find an alternative treatment that minimizes complications and maximizes efficacy. This study was conducted to investigate the role of rosmarinic acid on the neuropathic pain by studying the serotonin pathways in a model of neuropathic pain in rats. Materials and Methods: In this study, 66 Wistar rats weighing 200-250gr were used. Neuropathic pain was created by the spinal nerve ligation (SNL). Three groups of rats received intraperitoneal injection of a single dose of rosmarinic acid (10, 20 and 40 mg /kg) on day 7 to evaluate the acute effect of rosmarinic acid. In order to evaluate the role of serotonin in the antinociceptive effect of rosmarinic acid, a group of rats received serotonin discharger (PCPA) at a dose of 150 mg/kg intrapenitoneal on 5, 6 and 7 days. Serotonin receptor antagonists (5-HT1A, 5-HT2A, 5-HT1B, 5-HT3) with doses of 10, 35, 20, 30 μg/rat, respectively intrathecal was injected on day 7. Ten minutes after the injection of antagonists, rosmarinic acid 40mg/kg was administered to determine the effect of the receptors. The symptoms of neuropathic pain were evaluated through behavioral tests (thermal hayperalgesia and mechanical allodynia) on the seventh day. Results: Mechanical allodynia and thermal hyperalgesia in the SNL group received normal saline started on day 2 after surgery and reached the highest level on day 7, which was quite significant in comparison with the sham group. Intraperitoneal injection of rosmarinic acid 40 mg/kg/day reduced both mechanical allodynia and thermal hyperalgesia. Intraperitoneal injection of PCPA with rosmarin acid increased the behavioral symptoms of pain compared with that of animals received rosmarin acid (40 mg/kg). Intratechal injection of 5-HT1A, 5-HT2A, 5-HT1B, 5-HT3 receptor antagonists with rosmarinic acid (40 mg/kg), on day 7, reveresed mechanical anti-allodynic and thermal anti-hyperalgesic effects of rosmarinic acid. Conclusion: Our study suggests that rosmarinic acid at a dose of 40 mg/kg can have acute anti-allodynic and hyperalgesic effects following the induction of neuropathic pain. Our observations also indicate serotonergic pathway involvement in the anti-nociceptive effects of rosmarinic acid. Additionally, the intracton of rosmarinic acid with 5-HT1A, 5-HT1B, 5-HT2A and 5-HT3 receptors could at least in part responsible for the antinociceptive effect of rosmarinic acid.
